- Seminar Topic: Super-Resolution Fluorescence Microscopy: Technology Development and Biological Application Research
- Seminar Overview:
Single Molecule Localization Microscopy (SMLM) has transformed optical microscopy by achieving spatial resolutions as fine as 20 nm, enabling new insights into nanoscale structures in cells and their physiological functions. Awarded the 2014 Nobel Prize in Chemistry, this super-resolution technique has extended our understanding of subcellular organization. In this seminar, I will introduce SMLM's foundational principles and our lab's adaptation, oblique plane SMLM (obSTORM), which applies to a broader range of biological samples, including tissues and small animals. I will also present recent efforts to improve obSTORM's imaging performance through the development of a high refractive-index imaging buffer, advanced point spread function modeling, and deep learning-based background fluorescence removal. Additionally, I will briefly cover ongoing biological application studies in our lab using SMLM. SMLM offers new opportunities for nanoscale biological research and advances in medicine.
